Effects of UE on colchicine and on human cancer cells

Cells were stained by H+E for this study.

When native colchicine was applied on cancer cells, without any UE transfer to either the product nor the cells (A), approximately 70% of cells were affected. When the cells were treated with UE, which promotes their elimination, even when only inactivated colchicine was added to them (B), the rate of effectiveness jumped to 95% (inactivation of the colchicine diminishes its toxicity by approx. 40%).

(C) shows the effects of inactivated colchicine on cancer cells which did not get any UE transfers. There is about 40% toxicity left after colchicine inactivation.

When native colchicine was applied on cancer cells (A), it induced apoptosis. This picture shows many cells expressing the characteristic of positive fluorescent staining, which indicates that many cells are apoptosis. When the colchicine was inactivated by UE, a clear decrease in apoptosis expression was seen (B), close to the control (C), which did not receive any colchicine application.

Cancer cells without any UE treatment proliferated and expressed no or few apoptosis cells.

H+E stained reconstituted human epidermis (RHE) tissue sections

This figure shows a comparison between a UE-treated RHE and an untreated RHE on day 11 (d11). In the UE-treated RHE at low magnification 10X (A) and at higher magnification 40X (C), there is evidence of a very nice structure of epidermis with all its layers.

In the untreated RHE (B at 10X and D at 40X) there is slightly less keratin and epidermal thickness than in the treated one.

This figure shows a comparison between a UE-treated RHE and an untreated RHE on day 13 (d13).

The UE-treated RHE (A,C) shows well-structured, healthy, thick epidermis and keratin as a result of active cell growth.

The untreated RHE (B,D) shows less thickness of epidermis and keratin, resulting in a less developed structure.
